Various virulence determinants in Pseudomonas aeruginosa are regulated by the quorum sensing (QS) network producing and releasing signalling molecules. Two of these virulence determinants are the pyocyanin and pyoverdine, which interfere with multiple cellular functions during infection. The application of QS-inhibiting agents, such as cyclodextrins (CDs), appears to be a promising approach. Further to method development, this research tested in large-volume test systems the effect of α- and ß-CD (ACD, BCD) at 1, 5, and 10 mM concentrations on the production of pyocyanin in the P. aeruginosa model system. The concentration and time-dependent quorum quenching effect of native CDs and their derivatives on pyoverdine production was tested in a small-volume high-throughput system. In the large-volume system, both ACD and BCD significantly inhibited pyocyanin production, but ACD to a greater extent. 10 mM ACD resulted in 58% inhibition, while BCD only ~40%. Similarly, ACD was more effective in the inhibition of pyoverdine production; nevertheless, the results of RMANOVA demonstrated the significant efficiency of both ACD and BCD, as well as their derivatives. Both the contact time and the cyclodextrin treatments significantly influenced pyoverdine production. In this case, the inhibitory effect of ACD after 48 h at 12.5 mM was 57%, while the inhibitory effect of BCD and its derivatives was lower than 40%. The high-level significant inhibition of both pyocyanin and pyoverdine production by ACD was detectable. Consequently, the potential value of CDs as QS inhibitors and the antivirulence strategy should be considered. KEYPOINTS: • Applicability of a simplified method for quantification of pyocyanin production was demonstrated. • The cyclodextrins significantly affected the pyocyanin and pyoverdine production. • The native ACD exhibited the highest attenuation in pyoverdine production.

The performance of combined reduction of nitrate (NO3 - ) to dinitrogen gas (N2 ) and oxidation of arsenite (As[III]) to arsenate (As[V]) by a bioelectrochemical system was assessed, supported by ecotoxicity characterization. For the comprehensive toxicity characterization of the untreated model groundwater and the treated reactor effluents, a problem-specific ecotoxicity test battery was established. The performance of the applied technology in terms of toxicity and target pollutant elimination was compared and analyzed. The highest toxicity attenuation was achieved under continuous flow mode with hydraulic retention time (HRT) = 7.5 h, with 95%, nitrate removal rate and complete oxidation of arsenite to arsenate. Daphnia magna proved to be the most sensitive test organism. The results of the D. magna lethality test supported the choice of the ideal operational conditions based on chemical data analysis. The outcomes of the study demonstrated that the applied technology was able to improve the groundwater quality in terms of both chemical and ecotoxicological characteristics. The importance of ecotoxicity evaluation was also highlighted, given that significant target contaminant elimination did not necessarily lower the environmental impact of the initial, untreated medium, in addition, anomalies might occur during the technology operational process which in some instances, could result in elevated toxicity levels.

After more than a decade of studying the ecotoxicity of graphene oxide nanomaterials (nGOs), it has been concluded that there is limited information available regarding the environmental risk of graphene-based materials. Since existing ecotoxicological studies of nanomaterials have produced contradictory results, it is recommended that case-by-case studies should be conducted to evaluate their effects. This can be carried out by employing several methods, testing species from different trophic levels, and conducting community studies. Our goal was to evaluate the toxicity effects of two GOs (AF 96/97 and PM 995) derived from different graphite precursors on various test organisms from diverse trophic levels (bacteria, protozoa, a freshwater microbial community, plants, and invertebrate animals) in aquatic environments. We compared the effects of both nGO types and estimated the predicted no-effect environmental concentration (PNEC) values to determine their potential environmental risk. Our findings demonstrated the need for a complex ecotoxicity toolkit since the ecotoxicity results varied based on the test organism, the selected endpoints, and the test method used. Additionally, we found that toxicity effects were dependent on the concentration and characteristics of the specific nGO type used, as well as the exposure time. We estimated the PNEC values for GO AF 96/97 and GO PM 995 in the aquatic compartment to be 8 ng/L and 4 ng/L, respectively. Even after applying the worst-case scenario approach, the tested nGOs pose no environmental risk.

OBJECTIVES: The applicability of a simple and high-throughput method for quantitative characterization of biofilm formation by Candida boidinii was tested in order to evaluate the effects of exogenous tyrosol on yeast growth and biofilm formation capacity. RESULTS: Significant concentration-, temperature and time-dependent effect of tyrosol (2-(4-hydroxyphenyl)ethanol) was demonstrated, but it differentially affected the growth and biofilm formation (characterized by crystal violet staining and XTT-reduction assay) of Candida boidinii. Testing biofilm based on metabolic activity displayed sensitively the differences in the intensity of biofilm in terms of temperature, tyrosol concentration, and exposure time. At 22 °C after 24 h none of the tyrosol concentrations had significant effect, while at 30 °C tyrosol-mediated inhibition was observed at 50 mM and 100 mM concentration. After 48 h and 72 h at 22 °C, biofilm formation was stimulated at 6.25-25 mM concentrations, meanwhile at 30 °C tyrosol decreased the biofilm metabolic activity proportionally with the concentration. CONCLUSIONS: The research concludes that exogenous tyrosol exerts unusual effects on Candida boidinii growth and biofilm formation ability and predicts its potential application as a regulating factor of various fermentations by Candida boidinii.

The lack of high-grade scandium (Sc) ores and recovery strategies has stimulated research on the exploitation of non-ore-related secondary sources that have great potential to safeguard the critical raw materials supply of the EU's economy. Waste materials may satisfy the growing global Sc demand, specifically residues from titanium dioxide (TiO2) production. New technologies are being developed for the recovery of Sc from such residues; however, the possible environmental impacts of intermediary products and residues are usually not considered. In order to provide a comprehensive ecotoxicity characterisation of the wastes and intermediate residues resulting from one promising new technology, acid-resistant nanofiltration (arNF), a waste-specific ecotoxicity toolkit was established. Three ecotoxicity assays were selected with specific test parameters providing the most diverse outcome for toxicity characterisation at different trophic levels: Aliivibrio fischeri (bacteria) bioluminescence inhibition (30 min exposure), Daphnia magna (crustacean) lethality and immobilisation (24 h exposure) and Lemna minor (plant) growth inhibition with determination of the frond number (7 d exposure). According to our results, the environmental impact of the generated intermediate and final residues on the aquatic ecosystem was mitigated by the consecutive steps of the filtration methods applied. High and statistically significant toxicity attenuation was achieved according to each test organism: toxicity was lowered based on EC20 values, according to the A. fischeri bioluminescence inhibition assay (by 97%), D. magna lethality (by 99%) and L. minor frond number (by 100%), respectively, after the final filtration step, nanofiltration, in comparison to the original waste. Our results underline the importance of assessing chemical technologies' ecotoxicological and environmental impacts with easy-to-apply and cost-effective test methods to showcase the best available technologies.

Aim: Although many children who experience ischemic stroke come from bilingual backgrounds, it is unclear whether bilingual exposure affects poststroke development. Our research evaluates bilingual and monolingual exposure on linguistic/cognitive development poststroke across 3 stroke-onset groups. Method: An institutional stroke registry and medical charts were used to gather data on 237 children across 3 stroke-onset groups: neonatal, <28 days; first-year, 28 days to 12 months; and childhood, 13 months to 18 years. The Pediatric Stroke Outcome Measure (PSOM), administered several times poststroke, was used to evaluate cognition and linguistic development. Results: Similar cognitive outcomes were observed across language groups. However, an interaction effect with stroke-onset group was observed, with monolinguals in the first-year group having worse productive language outcomes as compared to bilinguals. Interpretation: Overall, no detrimental effects of bilingualism were found on children's poststroke cognition and linguistic development. Our study suggests that a bilingual environment may facilitate language development in children poststroke.

Heterogeneous photocatalysis has been increasingly investigated during the past years and has been recognized as a promising technique for clean and safe water purification. The current study exploits the advantage of this technique demonstrating that the removal of a biorefractory water pollutant named clofibric acid can be really improved by photocatalysis through a parametric comprehensive investigation and optimization study based on response surface methodology. Its novelty comes from the approach used to enhance the efficiency of the photocatalytic degradation of clofibric acid. A custom central composite design consisting of 49 trials was applied for process modeling and a quadratic robust model was derived based on the analysis of variance for the optimization of the process parameters. The effective removal of the target molecule with about 70% carbon mineralization was achieved under optimal photocatalytic conditions: 1.5 mg/L as the initial concentration of pollutant, 0.61 g/L catalyst, and an irradiation time of 190 min. Further, it was provided that nitrates play a positive role in the removal of this pollutant, while hydrogenocarbonates slow down its elimination. The ecotoxicity evaluation at different trophic levels confirmed the low toxicity of photodegradation by-products. Data analysis demonstrated that response surface methodology is a reliable approach for the optimization of the interactive effects of photocatalytic process parameters and is able to enhance their performance for the complete elimination of this hardly removed water pollutant.

Electrobioremediation technologies hold considerable potential for the treatment of soils contaminated by petroleum hydrocarbons (PH), since they allow stimulating biodegradation processes with no need for subsurface chemicals injection and with little to no energy consumption. Here, a microbial electrochemical snorkel (MES) was applied for the treatment of a soil contaminated by hydrocarbons. The MES consists of direct coupling of a microbial anode with a cathode, being a single conductive, non-polarized material positioned suitably to create an electrochemical connection between the anoxic zone (the contaminated soil) and the oxic zone (the overlying oxygenated water). Soil was also supplemented with electrically conductive particles of biochar as a strategy to construct a conductive network with microbes in the soil matrix, thus extending the radius of influence of the snorkel. The results of a comprehensive suite of chemical, microbiological and ecotoxicological analyses evidenced that biochar addition, rather than the presence of a snorkel, was the determining factor in accelerating PH removal from contaminated soils, possibly accelerating syntrophic and/or cooperative metabolisms involved in the degradation of PH. The enhancement of biodegradation was mirrored by an increased abundance of anaerobic and aerobic microorganisms known to be involved in the degradation of PH and related functional genes. Plant ecotoxicity assays confirmed a reduction of soils toxicity in treatments receiving electrically conductive biochar.

Quorum sensing (QS) is a population-density-dependent communication process of microorganisms to coordinate their activities by producing and detecting low-molecular-weight signal molecules. In pathogenic bacteria, the property controlled by QS is often related to infectivity, e.g., biofilm formation. Molecular encapsulation of the QS signals is an innovative method to prevent the signals binding to the receptors and to attenuate QS. Cyclodextrins (CDs) may form an inclusion complex with the signals, thus reducing the communication (quorum quenching, QQ). A systematic study was performed with α-, ß-cyclodextrin, and their random methylated, quaternary amino and polymer derivatives to evaluate and compare their effects on the biofilm formation of Pseudomonas aeruginosa. To examine the concentration-, temperature- and time-dependency of the QQ effect, the CDs were applied at a 0.1-12.5 mM concentration range, and biofilm formation was studied after 6, 24, 48 and 72 h at 22 and 30 °C. According to the results, the QS mechanism was significantly inhibited; the size of the cavity, the structure of the substituents, as well as the monomeric or polymeric character together with the concentration of the CDs have been identified as key influencing factors of biofilm formation. Statistically determined effective concentration values demonstrated outstanding efficiency (higher than 80% inhibition) of α-CD and its random methylated and polymer derivatives both on the short and long term. In summary, the potential value of CDs as inhibitors of QS should be considered since the inhibition of biofilm formation could significantly impact human health and the environment.

Spontaneous, task-free, hemodynamic activity of the brain provides useful information about its functional organization, as it can describe how different brain regions communicate to each other. Neuroimaging studies measuring the spontaneous activity of the brain are conducted while the participants are not engaged in a particular task or receiving any external stimulation. This approach is particularly useful in developmental populations as brain activity can be measured without the need for infant compliance and the risks of data contamination due to motion artifacts. In this project we sought to i) characterize the intrinsic functional organization of the brain in 4-month-old infants and ii) investigate whether bilingualism, as a specific environmental factor, could lead to adaptations on functional brain network development at this early age. Measures of spontaneous hemodynamic activity were acquired in 4-month-old infants (n = 104) during natural sleep using functional near-infrared spectroscopy (fNIRS). Emphasis was placed on acquiring high-quality data that could lead to reproducible results and serve as a valuable resource for researchers investigating the developing functional connectome.

OBJECTIVES: Previous virtual care literature within the field of speech-language pathology has primarily focused on validating the virtual use of intervention programmes. There are fewer articles addressing the validity of conducting virtual assessments, particularly standardized assessment of oral language and literacy abilities in children. In addition, there is a lack of practical, useful recommendations available to support clinicians and researchers on how to conduct these assessment measures virtually. Given the recent rapid rise in virtual care and research as a result of the Coronavirus-19 pandemic, clinicians and researchers require guidance on best practices for virtual administration of these tools imminently. This article seeks to fill this gap in the literature by providing such recommendations. METHODS: We (a) completed a narrative review of the extant literature, and (b) conducted semi-structured interviews with a group of 12 clinicians, students and researchers who had administered standardized language and literacy assessments with a variety of monolingual and multilingual school-aged children, with and without speech and language difficulties, in clinical and research settings. Six themes: candidacy for virtual assessment, communication and collaboration with caregivers, technology and equipment, virtual administration, ethics, consent and confidentiality, and considerations for bilingual populations were identified as a result of these two processes and were used to develop a set of recommendations to guide the use of standardized assessments in a virtual setting. In line with the Guidelines International Network, these recommendations were rated by group members, and reviewed by external stakeholders. A quasi-Delphi consensus procedure was used to reach agreement on ratings for recommendations. RESULTS: We have developed and outlined several recommendations for clinicians and researchers to guide their use of standardized language and literacy assessments in virtual care, across six key themes. CONCLUSIONS: This article is one of the first to share practical recommendations for virtual assessment in the domain of oral language and literacy assessment for clinicians and researchers. We hope the current recommendations will facilitate future clinical research in this area, and as the body of research in this field grows, this article will act as a basis for the development of formal Clinical Practice Guidelines.

An extensive literature exists regarding the effect of bilingualism on cognition in developing populations. However, the term 'cognition' is vague and applies to a large number of different abilities. We reviewed 60 publications examining cognition in simultaneous bilingual children to understand what aspects of cognition have been studied in this population and what tasks have been used, in addition to qualitatively assessing the results of bilingual/monolingual comparisons. Executive function was the most frequently assessed cognitive ability across all age groups, paralleling the adult bilingual literature, with memory flexibility and theory of mind also emerging as common targets within infant and preschool age groups. Results are discussed in light of developmental trajectories and assessment methodologies currently available for the cognitive abilities represented in this literature.

The application of Biolog EcoPlate™ for community-level physiological profiling of soils is well documented; however, the functional diversity of aquatic bacterial communities has been hardly studied. The objective of this study was to investigate the applicability of the Biolog EcoPlate™ technique and evaluate comparatively the applied endpoints, for the characterisation of the effects of metal oxide nanoparticles (MONPs) on freshwater microbial communities. Microcosm experiments were run to assess the effect of nano ZnO and nano TiO2 in freshwater at 0.8-100 mg/L concentration range. The average well colour development, substrate average well colour development, substrate richness, Shannon index and evenness, Simpson index, McIntosh index and Gini coefficient were determined to quantify the metabolic capabilities and functional diversity. Comprehensive analysis of the experimental data demonstrated that short-term exposure to TiO2 and ZnO NPs affected the metabolic activity at different extent and through different mechanisms of action. TiO2 NPs displayed lower impact on the metabolic profile showing up to 30% inhibition. However, the inhibitory effect of ZnO NPs reached 99% with clearly concentration-dependent responses. This study demonstrated that the McIntosh and Gini coefficients were well applicable and sensitive diversity indices. The parallel use of general metabolic capabilities and functional diversity indices may improve the output information of the ecological studies on microbial communities.

Behavioral studies examining vowel perception in infancy indicate that, for many vowel contrasts, the ease of discrimination changes depending on the order of stimulus presentation, regardless of the language from which the contrast is drawn and the ambient language that infants have experienced. By adulthood, linguistic experience has altered vowel perception; analogous asymmetries are observed for non-native contrasts but are mitigated for native contrasts. Although these directional effects are well documented behaviorally, the brain mechanisms underlying them are poorly understood. In the present study we begin to address this gap. We first review recent behavioral work which shows that vowel perception asymmetries derive from phonetic encoding strategies, rather than general auditory processes. Two existing theoretical models-the Natural Referent Vowel framework and the Native Language Magnet model-are invoked as a means of interpreting these findings. Then we present the results of a neurophysiological study which builds on this prior work. Using event-related brain potentials, we first measured and assessed the mismatch negativity response (MMN, a passive neurophysiological index of auditory change detection) in English and French native-speaking adults to synthetic vowels that either spanned two different phonetic categories (/y/vs./u/) or fell within the same category (/u/). Stimulus presentation was organized such that each vowel was presented as standard and as deviant in different blocks. The vowels were presented with a long (1,600-ms) inter-stimulus interval to restrict access to short-term memory traces and tap into a "phonetic mode" of processing. MMN analyses revealed weak asymmetry effects regardless of the (i) vowel contrast, (ii) language group, and (iii) MMN time window. Then, we conducted time-frequency analyses of the standard epochs for each vowel. In contrast to the MMN analysis, time-frequency analysis revealed significant differences in brain oscillations in the theta band (4-8 Hz), which have been linked to attention and processing efficiency. Collectively, these findings suggest that early-latency (pre-attentive) mismatch responses may not be a strong neurophysiological correlate of asymmetric behavioral vowel discrimination. Rather, asymmetries may reflect differences in neural processing efficiency for vowels with certain inherent acoustic-phonetic properties, as revealed by theta oscillatory activity.

Significance: Early monolingual versus bilingual experience induces adaptations in the development of linguistic and cognitive processes, and it modulates functional activation patterns during the first months of life. Resting-state functional connectivity (RSFC) is a convenient approach to study the functional organization of the infant brain. RSFC can be measured in infants during natural sleep, and it allows to simultaneously investigate various functional systems. Adaptations have been observed in RSFC due to a lifelong bilingual experience. Investigating whether bilingualism-induced adaptations in RSFC begin to emerge early in development has important implications for our understanding of how the infant brain's organization can be shaped by early environmental factors. Aims: We attempt to describe RSFC using functional near-infrared spectroscopy (fNIRS) and to examine whether it adapts to early monolingual versus bilingual environments. We also present an fNIRS data preprocessing and analysis pipeline that can be used to reliably characterize RSFC in development and to reduce false positives and flawed results interpretations. Methods: We measured spontaneous hemodynamic brain activity in a large cohort ( N = 99 ) of 4-month-old monolingual and bilingual infants using fNIRS. We implemented group-level approaches based on independent component analysis to examine RSFC, while providing proper control for physiological confounds and multiple comparisons. Results: At the group level, we describe the functional organization of the 4-month-old infant brain in large-scale cortical networks. Unbiased group-level comparisons revealed no differences in RSFC between monolingual and bilingual infants at this age. Conclusions: High-quality fNIRS data provide a means to reliably describe RSFC patterns in the infant brain. The proposed group-level RSFC analyses allow to assess differences in RSFC across experimental conditions. An effect of early bilingual experience in RSFC was not observed, suggesting that adaptations might only emerge during explicit linguistic tasks, or at a later point in development.

We assessed monolingual Spanish and bilingual Spanish-Basque toddlers' sensitivity to gender agreement in correct vs. incorrect Spanish noun phrases (definite article + noun), using a spontaneous preference listening paradigm. Monolingual Spanish-learning toddlers exhibited a tendency to listen longer to the grammatically correct phrases (e.g., la casa; "the house"), as opposed to the incorrect ones (e.g., *el casa). This listening preference toward correct phrases is in line with earlier results obtained from French monolingual 18-month-olds (van Heugten & Christophe, 2015). Bilingual toddlers in the current study, however, tended to listen longer to the incorrect phrases. Basque was not a source of interference in the bilingual toddler's input as Basque does not instantiate grammatical gender agreement. Overall, our results suggest that both monolingual and bilingual toddlers can distinguish between the correct and incorrect phrases by 18 months of age; however, monolinguals and bilinguals allocate their attention differently when processing grammatically incorrect forms.

RESUMO

Bacterial Quorum Sensing is a cell-to-cell communication process, in which, bacteria, performing cooperative behaviour, produce and detect extracellular signalling chemicals, to monitor cell population density. Numerous bacterial processes including bioluminescence, virulence factor production, biofilm formation etc. are known to be influenced by this bacterial communication network. Interest in QS systems has emerged in response to the fact that these processes have significant impact on the environment, human health as well as agriculture. Cyclodextrins-mediated quorum quenching is an innovative approach and the available information about their effects is very scarce. We selected Aliivibrio fischeri, a bacterium, producing light, based on Quorum Sensing, to be the first to investigate the cyclodextrins' effect on this bioluminescence. A systematic study was performed with twelve different cyclodextrin compounds in order to determine their concentration- and time-dependent bioluminescence inhibitory effect in the A. fischeri model system. Especially high quorum quenching effect was found for α-cyclodextrin: 10 mM α-cyclodextrin at 120 min contact time which caused ~64% inhibition of bioluminescence. Experiments with the co-administration of α-cyclodextrin and N-(3-oxohexanoyl)-L-homoserine lactone, the signalling molecule of A. fischeri clearly showed, that the stimulating effect of this signal was diminished by α-cyclodextrin, suggesting, that complexation was responsible for the observed Quorum Sensing suppression. Although ß-cyclodextrin and its hydroxypropyl derivative significantly inhibited bioluminescence at as low as 0.156 mM concentration, their efficiency did not reach the level of α-cyclodextrin. According to our results, the autoinducer-dependent quorum sensing mechanism in Aliivibrio fischeri was markedly inhibited, the quorum quenching effect of cyclodextrins was clearly demonstrated. The efficiency was influenced by several parameters; the size of the interior cavity, the structure and the concentration of the cyclodextrins, as well as the contact time with the cells. The application of a cyclodextrin-trap for complexation of signal molecules may be a novel, promising method for influencing QS interfering strategies, for example, to enhance the efficiency of various biotechnologies, as well as to find alternative approaches against bacterial proliferation and infections. Furthermore, our results could also serve as a basis for further research with bacterial or plant model systems, in which the same chemical signals may induce physiological responses.

The extensive use of engineered nanomaterials, such as graphene oxide (GO), is stimulating research about its potential environmental impacts on the aquatic ecosystem. This study is aimed to comprehensively assess the acute toxicity of a well-characterized GO suspension to Daphnia magna. Conventional ecotoxicological endpoints (lethality, immobilization) and more sensitive, sublethal endpoints (heartbeat rate, feeding activity, and reactive oxygen species (ROS)) production were used. The possible normalization of the heartbeat rate and feeding activity in clean test medium was also investigated. The fate, time-dependent, and concentration-dependent aggregation behaviour of GO was followed by dynamic light scattering, UV-Vis spectroscopy, and zeta potential measurement methods. The EC20 value for immobilization was 50 mg/L, while, for physiological and behavioural endpoints, it ranged from 8.1 mg/L (feeding activity) to 14.8 mg/L (immobilization). The most sensitive endpoint was the ROS production with EC20 = 4.78 mg/L. 24-h recovery experiments revealed that feeding activity was restored only up to a certain level at higher concentrations, indicating that the potential environmental health effects of GO cannot be neglected. Alterations of normal physiology (heart rate) and feeding activity may be associated with increased risk of predation and reproductive decline, highlighting that GO may have impacts on population and food web dynamics in aquatic ecosystems.

Attunement theories of speech perception development suggest that native-language exposure is one of the main factors shaping infants' phonemic discrimination capacity within the second half of their first year. Here, we focus on the role of acoustic-perceptual salience and language-specific experience by assessing the discrimination of acoustically subtle Basque sibilant contrasts. We used the infant-controlled version of the habituation procedure to assess discrimination in 6- to 7-month and 11- to 12-month-old infants who varied in their amount of exposure to Basque and Spanish. We observed no significant variation in the infants' discrimination behavior as a function of their linguistic experience. Infants in both age-groups exhibited poor discrimination, consistent with Basque adults finding these contrasts more difficult than some others. Our findings are in agreement with previous research showing that perceptual discrimination of subtle speech sound contrasts may follow a different developmental trajectory, where increased native-language exposure seems to be a requisite.
